Charismatic Polish opera singer Jan Kiepura made his Hollywood debut in Give Us This Night. His thick Slavic accent notwithstanding, Kiepura is cast as Italian fisherman Antonio. In the habit of singing as he fishes, Antonio catches the attention of opera diva Maria (played by real-life operatic soprano Gladys Swarthout, in her second film appearance). Herbert Lanyon is thought to be dead after a shipwreck, and his fiancée Mildred Carrison is forced by her money-minded Aunt Clara into marriage with "Bull" Magee, a gambler and underworld boss who mistreats Mildred. After Herbert returns, Magee undergoes financial difficulties that he blames on Mildred and Herbert, and seeks revenge. Herbert and a repentant Aunt Clara, however, free Mildred from Magee, and the lovers are able to marry.

Sam Johnson & Raskus Hudson are suitors for Mandy Jones. When Raskus Bug get's the 'cold shoulder', his negro blood is aroused. Raskus & his gang steal a chicken and plant it in Sam's house, and put chicken feathers in Sam's shirt. Sam gets thrown into jail. Mandy gets Sherlock Jackson Holmes, the great detective of International fame, who finds a trail of chicken feathers which leads to the gang's den. The gang gets sent to the dungeon freeing the innocent Sam.

Mattie Cook, the undertaker's daughter, loves John Scott, who has no job. Her father wants her to marry Sime Sloan, who has a job, and it takes all of Mattie's persuasive power to overcome Dad's objections, but she is equal to the occasion. She gets rid of John's two rivals, Sime and Bime, by promising to marry them if they will prove their love for her. One must sleep in one of her father's coffins and the other sit by it all night. Unknown to one another they come to fulfill their promise.
